FDA Approves Donidalorsen to Prevent Hereditary Angioedema Attacks

On August 21, 2025, the FDA approved Ionis Pharmaceuticals’ donidalorsen (DAWNZERA), an RNA-targeted medicine, to prevent HAE attacks in adults and children aged 12 years or older. Semaglutide Reduces Medical Costs, Healthcare Resource Utilization in Heart Failure

Findings from a real-world analysis of Komodo Healthcare Map data suggest use of semaglutide 2.4 mg incurs lower medical costs and inpatient resource utilization among patients with overweight or obesity and heart failure compared with those who did not receive the GLP-1 receptor agonist. Specifically, semaglutide treatment was associated with 28% lower all-cause medical costs, 55% lower inpatient hospitalization costs, and 42% lower inpatient visit rate, a key driver of the lower costs observed with semaglutide.

RE104 Reduces Postpartum Depression Symptoms by Day 7 in Phase 2 RECONNECT

On August 19, 2025, Reunion Neuroscience announced the phase 2 RECONNECT trial met its primary endpoint, with RE104 providing a significant reduction in the Montgomery-Åsberg Depression Rating Scale (MADRS) from baseline to day 7 in patients with postpartum depression.

Dyslipidemia, Other Metabolic Syndrome Factors May Raise Parkinson’s Disease Risk

A larger waistline and higher blood pressure, among other factors connected to metabolic syndrome, may be associated with an increased risk of Parkinson disease, according to findings from a recent study. After adjusting for age, smoking status, physical activity and genes that increase the risk of Parkinson disease, investigators found people with metabolic syndrome were about 40% more likely to develop Parkinson’s disease than those without the syndrome.

Based on analysis of a nationally representative sample of US adults, 11.8 million are eligible for statin therapy with a 10-year 

atherosclerotic cardiovascular disease (ASCVD)

 risk ≥5% with the Predicting Risk of Cardiovascular Disease Events (PREVENT) equations.

Although the 2018 Cholesterol Guideline defines increased 10-year ASCVD risk as ≥7.5% based on pooled cohort equations (PCEs), recent data have suggested that PCEs overestimate risk substantially. To that end, the American Heart Association established the PREVENT equations, which provide clinicians with risk estimates for total cardiovascular disease, as well as ASCVD and 

. The equations therefore represent a single multivariable risk equation for an easily implemented and simplified framework.

“We examined population-level implications for statin eligibility according to different 10-year ASCVD risk estimates based on PREVENT and compared with guideline-defined thresholds based on PCEs,” wrote Sadiya Khan, MD, MSc, division of cardiology, Department of Medicine, Northwestern University Feinberg School of Medicine, and colleagues.

Investigators collected data from the National Health and Nutrition Examination Surveys, conducted from 2011-2020. They included nonpregnant individuals between the ages of 40-75 with available data for the PREVENT-ASCVD equations. Patients already taking statins and those eligible for statins due to comorbidities, such as ASCVD and diabetes, or cholesterol levels, such as a low-density lipoprotein C (LDL-C) count ≥190 mg/dL were excluded.

The team then calculated 10-year ASCVD risk for the remaining participants utilizing PREVENT and PCEs. 3 PREVENT-based ranges – 3%-3.9%, 4%-4.9%, and ≥5% - were evaluated based on estimated clinical benefit from statin trials. These were compared with PCE-based thresholds of borderline, intermediate, or high risk (5%-7.4% and ≥7.5%). Subgroup analyses were then conducted by age. Investigators calculated 10-year absolute risk reduction (ARR) assuming a 40% reduction in LDL-C with statins.

A total of 5242 participants, representing 133.6 million adults, 28.1 million were already taking statins, and 15.2 million were eligible for statins for secondary prevention or high-risk primary prevention due to diabetes or LDL-C ≥190 mg/dL. The remaining 70.2 million adults were divided into the 3 PREVENT-based 10-year ASCVD risk categories, with 7.9 million at 3-3.9%, 5.3 million at 4-4.9%, and 11.8 million at ≥5%.

Investigators noted that this translates to a mean 10-year ARR of 1.1%, 1.4%, and 2.2% for risk categories 3-3.9%, 4-4.9%, and ≥5%, respectively, if statins were recommended. Another 17 million adults were identified as having a 10-year ASCVD risk of≥7.5% with PCEs, making them eligible for statins, and 8.6 million may be considered for statins with a 10-year ASCVD risk of 5-7.4% with PCEs, with a mean 10-year ARR of 1.9% and 1.1%, respectively.

Khan and colleagues noted that the use of a PREVENT-based threshold of 4% or greater would result in an almost equivalent number of adults eligible for statins as the current guideline-based threshold of PCE 7.5% or greater. Additionally, patients with 10-year ASCVD risk of 3-3.9% and 4-4.9% had a 10-year ARR <2%, which suggests more marginal clinical benefits at these thresholds.

“However, given the safety and cost-effectiveness of statins, expanding eligibility may have the most favorable impact on ASCVD prevention,” Khan and colleagues wrote. “Ultimately, risk prediction is a first step in shared decision-making that incorporates risk thresholds, net benefit, and patient preferences.”

Validating Deucravacitinib’s Anticipated Expansion to Psoriatic Arthritis, with Philip Mease, MD

The psoriatic arthritis (PsA) field is looking forward to the potential addition of a new mechanism of action to the treatment landscape – deucravacitinib, an oral, selective TYK2 inhibitor already approved for people with psoriasis.

Based off of positive data from the phase 3 POETYK PsA-1 trial, the latest findings from which were presented at the

 European Alliance of Associations for Rheumatology (EULAR) Congress

 in June in Barcelona, Spain, Bristol Myers Squibb submitted and the FDA accepted the supplemental New Drug Application (sNDA) for the PsA indication with a Prescription Drug User Fee Act (PDUFA) goal date of March 6, 2026.

As presented at EULAR, POETYK PsA-1 met its primary endpoint of achieving ACR20 response at week 16 compared to placebo (54.2% vs 34.1%; 

<.0001) in adults with active PsA who were not previously treated with a biologic disease-modifying anti-rheumatic drug (bDMARD).The trial also met key secondary end points including Psoriasis Area and Severity Index (PASI) 75 response (51.9% vs 7.1%; 

 <.0001), Health Assessment Questionnaire-Disability Index (HAQ-DI) score (-.39 vs -.22; 

 <.0001), 36-Item Short Form Survey (SF-36) Physical Component Summary (PCS) score (6.06 vs 3.71; 

 <.0001) and Minimal Disease Activity (MDA) response (19.0% vs 10.2%; 

 spoke with study investigator Philip Mease, MD, director of rheumatology research at Providence Swedish Medical Center and clinical professor at the University of Washington School of Medicine, Seattle, to learn more about how the new data add to the field’s knowledge of deucravacitinib’s safety and efficacy for people with PsA and the potential the therapy may have in improving disease outcomes.

“It appears to have a more pristine safety profile, even [more so] than some of the JAK1 or JAK3 inhibitors. I think that patients will appreciate having a safe drug that can be used, especially early on, in PsA,” Mease said.

 in pediatric patients with atopic dermatitis in Sub-Saharan Africa—intensified by area-specific factors such as cultural stigma and socioeconomic challenges—suggests a greater quality of life burden compared to global statistics.

These findings were the result of a study conducted to examine the specific trends among children in Sub-Saharan Africa with this inflammatory skin disease. The research was authored by a team of investigators, including Edgar Mandeng Ma Linwa, from the Faculty of Health Sciences at the University of Buea in Cameroon.

“Using the PCC (Population, Concept and Context) framework in drafting a review question, this study aimed to answer the following question: What is the existing evidence on the QoL of pediatric patients with atopic dermatitis in Sub-Saharan Africa (SSA), including key influencing factors and research gaps?” Linwa and coauthors wrote.

Linwa and colleagues implemented the PRISMA-ScR framework, conducting a scoping review of studies published between 2010 and 2025 that explored atopic dermatitis in children and adolescents. The children the investigators assessed were aged 0–18 years and resided in in Sub-Saharan Africa.

Using the Embase, MEDLINE, APA PsycINFO, Google Scholar, ProQuest, African Journals Online (AJOL), and the Health Sciences and Diseases journal, the investigative team reviewed available literature. They sought to capture both high-quality international research and regionally relevant publications. The team specifically prioritized data from PubMed, Embase, PsycINFO, and ProQuest for peer-reviewed sources.

Linwa et al used Google Scholar to access to a broader range of region-specific research. Both Health Sciences and Diseases and AJOL were utilized due to their open access and their focus on Sub-Saharan Africa-based research. Eligible data related to sociodemographic characteristics, trial designs, quality of life outcomes, and atopic dermatitis severity. The information was synthesized by the investigative team via a biopsychological model and narrative approach.

Qualitative, cross-sectional, case-control, cohort, and review articles were considered by the team, whereas case reports, case series, and editorials were not. The data included in this analysis was required to be in English or French and to be released between 2010 - 2025, reflecting the primary scientific reporting languages used in Sub-Saharan Africa. Six trials met Linwa and coauthors' criteria: 5 cross-sectional studies and a single randomized controlled trial (RCT). These 6 studies were conducted in Côte d’Ivoire, Cameroon, South Africa, Madagascar, and Nigeria.

The numbers of trial subjects in these 6 trials ranged from 53 - 381, and they were primarily in urban hospital settings. Findings highlighted regional challenges, most notably the disproportionately high prevalence of non-mild AD.

Across these studies, up to 61.3% of pediatric patients presented with moderate disease and 24.2% with severe disease—figures which were noted to far exceed global estimates.

 Global trends suggest that severe atopic dermatitis is reported in fewer than 15% of cases. The investigators found a consistent link between burden of moderate to severe quality of life impairment to intense itch, disturbed sleep, and psychosocial stress, with SCORAD-assessed symptom severity and younger age (<1 year) being key correlates.

Quality evaluation via the Mixed Methods Appraisal Tool (MMAT) demonstrated to Linwa et al that methodological concerns, including reliance on urban-based samples along with insufficient reporting of nonresponse bias, diagnostic definition variations, and the use of instruments validated internationally but not culturally adapted for Sub-Saharan Africa. Socioeconomic status and comorbidities such as asthma were not frequently addressed, though both were highlighted by the team as likely to worsen the impacts of the disease.

In summary, this review of six Sub-Saharan Africa studies demonstrates that children with atopic dermatitis in the region frequently experience moderate to severe impairment to life quality. Unlike global patterns, where severe atopic dermatitis remains below 15%, they found that region-specific drivers contribute to a disproportionate disease and quality of life burdens in affected populations aged 0 - 18 years.

“The limited representation of only five SSA countries highlights significant research gaps, particularly in rural and East African settings, where cultural contexts may further influence QoL,” the investigators concluded.

 “Future research should prioritise rural, community-based studies using validated, context-specific QoL tools to capture SSA's diverse populations, employing mixed-method approaches to deepen insights into AD's multidimensional impact.”

Unique Challenges Highlighted in Pediatric Atopic Dermatitis in Sub-Saharan Africa

Investigators at Massachusetts General Hospital validated a 

history tool (DAHT) in a study of 79 participants.

“The [drug allergy history tool] contains survey items across key domains of the drug allergy history: medication exposure history, timing of reaction, signs and symptoms of reaction, and treatment of reaction,” wrote Kimberly G. Blumenthal MD, MSc, from the division of rheumatology, allergy, and immunology at Massachusetts General Hospital, and colleagues.

When it comes to drug allergy evaluations, having access to reaction history is critical. Electronic health records can be helpful, creating uniformity in data entry and improving accessibility across healthcare providers. A study published in May 2024 found that information on a patient’s medical history correlated with a positive antibiotic allergy test result. Children with a history of asthma or food allergy had an increased risk of positive allergic testing (

However, allergy records may not always be accurate or complete. Despite drug allergy labels affecting approximately one-third of patients in the United States, only about 14% have true immunologically mediated hypersensitivity reactions.

This is often the case for patients with penicillin allergy; studies have shown that 90 – 95% of patients with documented penicillin allergies can safely receive these medications after proper evaluation. Unnecessary avoidance of penicillin can have substantial consequences, such as increased healthcare costs, longer hospital stays, increased risk of adverse events, and development of antimicrobial resistance.

Investigators recognized the lack of a validated survey instrument to collect drug allergy history from patients and sought to validate a tool informed by existing questionnaires, peer-reviewed literature, and the expertise of the United States Drug Allergy Registry Consortium.

 The team ensured the tool would work for penicillin allergy by including items used in allergist risk stratification and known predictors of confirmed penicillin allergy.

Investigators revised the drug allergy history tool after 3 rounds of cognitive testing and after assessing for reliability, quality, and validity.

 Test-retest comparisons evaluated cognitive testing, and a concordance analysis against electronic health record allergist documentation evaluated reliability and validity.

Participants, recruited from allergy clinics at the Massachusetts General Hospital, had 1 or more drug allergies. The sample included 79 individuals with a mean age of 49 years and a demographic breakdown of 85% females, 85% White, and 11% Hispanic.

In total, 50 participants had multiple drug allergy levels, and 29 had single drug allergy labels. The most common drug allergies were penicillin (77%), sulfonamides (32%), cephalosporins (15%), and nonsteroidal anti-inflammatory drugs (8%).

The drug allergy history tool achieved an acceptable reliability of 86%. The tool had a more complete allergy history than allergist documentation in the electronic health record, with lower median uncertainty (21% DAHT vs 79% electronic health record).

“The DAHT is a reliable and valid source of patient-reported drug allergy information,” investigators concluded.

 “This tool can be used in clinical care and clinical research to obtain standardized patient-reported drug allergy history."

In an accompanying editorial, author Ricardo D. Zwiener, MD, from the division of allergy and immunology at Hospital Universitario Austral, Buenos Aires, Argentina, outlined several challenges of the drug allergy history tool that may “temper enthusiasm.”

He listed the lack of confirmatory testing, recall bias for remote reactions, and hurdles with electronic health record integration. Zwiener also noted that the validation cohort lacked diversity.

“The DAHT marks a significant step forward in modernizing drug allergy evaluation by centering patient-reported data within a structured, standardized framework,” Zwiener wrote. 

“Blumenthal and colleagues offer not only a validated tool but a scalable model for rethinking how allergy histories are captured and applied in both clinical and research settings… To fully realize this promise, future efforts must focus on broader validation across populations and seamless [electronic health record] integration.”
